The Lysosomal Positioning of mTOR Regulator and Regulation of Autophagy
نویسندگان
چکیده
Lysosomes are key cellular organelles that play a role in catabolism by degrading extracellular and intracellular material. Specially, mechanisitic target of rapamycin complex 1(mTOR) localizes in its active form to the surface of lysosome, which regulates energy level, growth signal, and nutrient etc [1]. In addition, there are reports related to a tight crosstalk between mTORC1 activity and lysosomal function.

Interpretation of bafilomycin, pH neutralizing or protease inhibitor treatments in autophagic flux experiments
Recent publications showed that the kinase MTOR localizes to lysosomes and its activation depends on amino acids inside the lysosomal lumen, implying that autophagic protein degradation is a positive regulator of MTOR in this setting. Since decreased MTOR activity results in autophagy induction, drug treatments that block autolysosomal degradation (a commonly used technique to estimate autophag...
متن کاملEupafolin ameliorates lipopolysaccharide-induced cardiomyocyte autophagy via PI3K/AKT/mTOR signaling pathway
Objective(s): Eupafolin, a major active component of Eupatorium perfoliatum L., has anti-inflammatory and anti-oxidant properties. Lipopolysaccharide (LPS) is responsible for myocardial depression. A line of evidences revealed that LPS induces autophagy in cardiomyocytes injury. This study aims to evaluate the effects of eupafolin on LPS-induced cardiomyocyte autophagy...
متن کاملKnockdown of mitofilin inhibits autophagy and facilitates starvation-induced apoptosis in HeLa cells
Objective(s): Mitofilin contributes to the maintenance of mitochondrial structure and functions. This study was undertaken to determine the mechanisms underlying its regulation of apoptosis. Materials and Methods: Mitofilin was knockdowned by specific short hairpin RNA (shRNA) and the stable HeLa cell clone was selected. The autophagy a...
متن کاملLoss of C9orf72 Enhances Autophagic Activity via Deregulated mTOR and TFEB Signaling
The most common cause of the neurodegenerative diseases amyotrophic lateral sclerosis and frontotemporal dementia is a hexanucleotide repeat expansion in C9orf72. Here we report a study of the C9orf72 protein by examining the consequences of loss of C9orf72 functions. Deletion of one or both alleles of the C9orf72 gene in mice causes age-dependent lethality phenotypes. We demonstrate that C9orf...
متن کاملRole of autophagy associated with Helicobacter pylori CagA and VacA toxins in gastric cancer
Helicobacter pylori (H. pylori) is a gram-negative microaerophilic bacterium that has been introduced as a cause of mucosal inflammation and gastric cancer. The most important pathogenic factors are VacA and CagA, which are associated with increased disease severity in clinical strains. Autophagy is a protected lysosomal degradation pathway degrading cytoplasmic content and is important in host...
